In the process of obtaining target extracts from natural raw materials or complex systems, the choice of extraction method is crucial. Different extraction methods are based on distinct principles and characteristics, making them suitable for various raw materials and target compounds. The following sections will introduce several common extract-extraction methods, analyzing their underlying principles, application scenarios, as well as their respective advantages and disadvantages.

(1) Solvent Extraction Method

This is one of the commonly used extraction methods. Depending on the polarity and solubility of the extract, an appropriate solvent—such as ethanol, water, or acetone—is selected to soak, decoct, or reflux-extract the raw materials. For example, water is used to extract water-soluble components from traditional Chinese medicinal herbs, while ethanol is used to extract fat-soluble components.

(2) Steam Distillation

It is primarily used to extract components that are volatile and can be distilled along with steam without being destroyed, such as essential oils and certain small-molecule alkaloids. This method is suitable for extracting aromatic compounds from plant materials, such as rose essential oil and peppermint oil.

(3) Supercritical Fluid Extraction Method

Extraction is carried out using supercritical fluids—such as carbon dioxide—which, in their supercritical state, exhibit high diffusivity and strong solvating power. This method offers advantages including high extraction efficiency, excellent selectivity, and the absence of solvent residues. It is often employed for extracting heat-sensitive and easily oxidized compounds, such as natural vitamins and unsaturated fatty acids.

(4) Ultrasonic Extraction Method

Through the cavitation effect, mechanical vibration, and thermal effects of ultrasound, the diffusion of extracts from the raw material into the solvent is accelerated. Compared with traditional extraction methods, ultrasonic extraction offers advantages such as shorter extraction time, higher extraction yield, and simpler operation. It has been widely applied in the preparation of extracts from plants, animals, and microorganisms.

(5) Microwave-assisted extraction method

By leveraging the thermal and non-thermal effects of microwaves, this method causes cell rupture in the raw materials, thereby accelerating the release of active ingredients. This technique features rapid extraction speed, low energy consumption, and excellent selectivity, making it suitable for the extraction of a wide variety of natural raw materials.
